Description

Principle of the Assay: Human CA125/MUC16 ELISA Kit was based on standard sandwich enzyme-linked immunesorbent assay technology. A monoclonal antibody from mouse specific for CA125/MUC16 has been precoated onto 96-well plates. Standards(Expression system for standard: CHO; Immunogen sequence: M21005-Q21992) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CA125/MUC16 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human CA125/MUC16 amount of sample captured in plate.

Background/Introduction: CA-125, also known as mucin 16 or MUC16, is a protein that in humans is encoded by the MUC16 gene. MUC16 is a member of the mucin family glycoproteins. It is a component of the ocular surface, the respiratory tract and the female reproductive tract epithelia. Since MUC16 is highly glycosylated it creates a hydrophilic environment that acts as a lubricating barrier against foreign particles and infectious agents on the apical membrane of epithelial cells. Also, the cytoplasmic tail of MUC16 has been shown to interact with cytoskeleton by binding members of the ERM protein family.
